Profilin is a major actin binding protein found in all eukaryotes and encoded by a small ancient gene family in plants
1. Profilin promotes actin assembly on the barbed ends of growing F-actin filaments

4. Profilin plays a role in ADP to ATP exchange in actin monomers preparing actin for polymerization into F-actin

2. Profilin sequesters G-actin monomers in heterodimeric profilactin complexes

 

5. Profilin binds several other ligands such as membrane polyphospho-inositides, short proline rich stretches in proteins, and the ARP2/3 complex

3. Profilin promotes depolymerization of actin filaments by binding G-actin
